Imunodeficiências Humanas & Reconstituição Imunitária

O nosso objetivo é compreender a patogénese de imunodeficiências do sistema imunitário humano e identificar estratégias de reconstituição imunológica. Uma área de interesses dominante é a imunopatogénese da infeção HIV/SIDA. Em contraste com o HIV-1, a infeção pelo HIV-2 associa-se a um aparente equilíbrio com o hospedeiro, traduzindo-se na ausência de impacto significativo na mortalidade na maioria dos adultos infetados.

A compreensão dos mecanismos que determinam o curso distinto destas duas infeções constitui a principal modelo de investigação, uma vez que Portugal é o único país não-africano com uma prevalência significativa de HIV-2. As estratégias de reconstituição dos linfócitos T tornaram-se uma área crítica de investigação para a prática clínica. A nossa investigação foca-se no estudo do desenvolvimento dos linfócitos T no timo e na sua posterior manutenção, com ênfase na IL-7 como citocina homeostática essencial, em contextos clínicos diferentes que incluem imunodeficiências, doenças autoimunes e transplantação.

Integramos um centro de referência de imunodeficiências primárias envolvendo o Hospital de Santa Maria e a FMUL. Estas doenças raras representam modelos naturais únicos para o esclarecimento do funcionamento do sistema imunitário. Um diagnóstico atempado é essencial para o sucesso terapêutico e aconselhamento genético, constituindo um desafio para a Imunologistas Clínica.
