System overview

The Cytek Aurora CS full spectrum technology provides flexibility, enabling the use of a wide array of new fluorochrome combinations without reconfiguring the system for each application. It delivers high-resolution data at the single-cell level to resolve the most challenging cell populations, such as cells with high autofluorescence or low levels of expression of key biomarkers, regardless of assay complexity.

Sorting can be performed into two or six tubes with speeds up to 25,000 events/second. The system sorts by incorporating cells from the sample tube into a stream of sterile PBS. The stream is interrogated by the lasers at the flow cell and the system electronics keeps track of each cell as they pass through the laser and determines specific cells that meet the sort criteria. A transducer vibrates the stream and induces droplet formation, with cells in the stream being incorporated into the droplets. If a cell meets the sorting criteria and is in the last drop before the break off, the instrument will charge that drop. The charged droplet is then deflected into the proper collection tube by the charge plates. Different cell types can be sorted with the use of 70 μm and 100 μm nozzles. The fully integrated Class II Type A2 biosafety cabinet meets personal and product protection, reducing the risk of operator exposure, while sorting potentially hazardous samples (e.g human samples and other BSL-2 level samples).

  • 5 spatially separated lasers: Ultra-violet (355nm), Violet (405nm), Blue (488nm), Yellow Green (561nm) and Red (640nm)
  • 3 scatter channels: 488nm FSC, 405nm SSC, 488nm SSC
  • 64 fluorescent detectors:
    • UV detector: 16 channels with optimized bandwidth from 365-829nm
    • V detector: 16 channels with optimized bandwidth from 420-829nm
    • B detector: 14 channels with optimized bandwidth from 498-829nm
    • YG detector: 10 channels with optimized bandwidth from 567-829nm
    • R detector: 8 channels with optimized bandwidth from 652-829nm
  • 70 μm and 100 μm nozzles available
  • Up to 6-way sorting into 1.5 ml, 4 ml tubes or PCR tubes and 2-way sorting into 15 ml tubes
  • Sorting for 6-, 12-, 24-, 96-, PCR stripes and microscope slides

It shares the same optical system as the CYTEK Aurora in Lisbon site, allowing for the easy transfer of experiments from the analyzer to the cell sorter without the need to modify your panels.

If you need high parameter cell sorting, check also the BD FACSymphony S6 SE in Lisbon site.
